Abstract
BACKGROUND:Clonidineisanα2-adrenoceptoragonist,whichhasanalgesicproperties.However,theanalgesicefficacyofperioperativeclonidineremainsunclear.We,therefore,testedthehypothesisthatclonidinereducesbothpainscoresandcumulativeopioidconsumptionduringtheinitial72hoursafternoncardiacsurgery.
METHODS:Sixhundredtwenty-fourpatientsundergoingelectivenoncardiacsurgeryundergeneralandspinalanesthesiawereincludedinthissubstudyofthePeriOperativeISchemiaEvaluation-2trial.Patientswererandomlyassignedto0.2mgoralclonidineorplacebo2to4hoursbeforesurgery,followedby0.2mg/dtransdermalclonidinepatchorplacebopatch,whichwasmaintaineduntil72hoursaftersurgery.Postoperativepainscoresandopioidconsumptionwereassessedfor72hoursaftersurgery.
