TODAY今日发布JAHAMar17,:9(6)今日发布06篇(共计27篇)CATHETERCARDIOINTEEarlyRecent,Mar14,今日发布01篇JournalofClinicalUltrasoundEarlyRecent,Mar14,今日发布01篇RECOMMEND推荐阅读01幽门螺杆菌感染通过外显子介导的机制损害内皮功能JAHAresearch-articleXiujuanXia,LinfangZhang,etc.2小时前等1用户推荐阅读本文BackgroundEpidemiologicalstudieshavesuggestedanassociationbetweenHelicobacterpylori(Hpylori)infectionandatherosclerosisthroughundefinedmechanisms.Endothelialdysfunctioniscriticaltothedevelopmentofatherosclerosisandrelatedcardiovasculardiseases.ThepresentstudywasdesignedtotestthehypothesisthatHpyloriinfectionimpairesendothelialfunctionthroughexosome‐mediatedmechanisms.流行病学研究表明,幽门螺杆菌(H-pylori)感染与动脉粥样硬化之间存在着联系,其机制尚不明确。内皮功能障碍是动脉粥样硬化及相关心血管疾病发生发展的关键。本研究旨在验证幽门螺杆菌感染通过外显子介导的机制损害内皮功能的假设。MethodsandResultsYoungmaleandfemalepatients(18‐35yearsold)withandwithoutHpyloriinfectionwererecruitedtominimizethechanceofpotentialriskfactorsforendothelialdysfunctionforthestudy.Endothelium‐dependentflow‐mediatedvasodilatationofthebrachialarterywasevaluatedinthepatientsandcontrolsubjects.MouseinfectionmodelswithCagAHpylorifromagastriculcerpatientwerecreatedtodetermineifHpyloriinfection‐inducedendothelialdysfunctioncouldbereproducedinanimalmodels.Hpyloriinfectionsignificantlydecreasedendothelium‐dependentflow‐mediatedvasodilatationinyoungpatientsandsignificantlyattenuatedacetylcholine‐inducedendothelium‐dependentaorticrelaxationwithoutchangeinnitroglycerin‐inducedendothelium‐independentvascularrelaxationinmice.Hpylorieradicationsignificantlyimprovedendothelium‐dependentvasodilationinbothpatientsandmicewithHpyloriinfection.ExosomesfromconditionedmediaofhumangastricepithelialcellsculturedwithCagAHpyloriorserumexosomesfrompatientsandmicewithHpyloriinfectionsignificantlydecreasedendothelialfunctionswithdecreasedmigration,tubeformation,andproliferationinvitro.InhibitionofexosomesecretionwithGWeffectivelypreservedendothelialfunctioninmicewithHpyloriinfection.年轻男性和女性患者(18-35岁)有或没有幽门螺杆菌感染,以尽量减少潜在的内皮功能障碍危险因素的机会为研究对象。评估患者和对照组肱动脉内皮依赖性血流介导的血管舒张功能。建立了胃溃疡患者CagAH-pylori小鼠感染模型,以确定H-pylori感染诱导的内皮功能障碍是否能在动物模型中复制。幽门螺杆菌感染显著降低年轻患者内皮依赖性血流介导的血管舒张,显著减弱乙酰胆碱诱导的内皮依赖性主动脉舒张,而不改变小鼠硝酸甘油诱导的内皮依赖性血管舒张。根除幽门螺杆菌可显著改善幽门螺杆菌感染患者和小鼠的内皮依赖性血管舒张功能。CagAH-pylori培养的人胃上皮细胞条件培养液外显子体或H-pylori感染患者和小鼠血清外显子体在体外显著降低内皮功能,降低移行、成管和增殖。GW抑制幽门螺杆菌感染小鼠外显子分泌,有效地保护了小鼠的内皮功能。ConclusionsHpyloriinfectionimpairedendothelialfunctioninpatientsandmicethroughexosome‐medicatedmechanisms.ThefindingsindicatedthatHpyloriinfectionmightbeanovelriskfactorforcardiovasculardiseases.幽门螺杆菌感染通过外源性药物机制损害患者和小鼠的内皮功能。提示幽门螺杆菌感染可能是心血管疾病的一个新的危险因素。扫描
